Lipoxygenase Pathway
5-Lipoxygenase
The enzyme 5-lipoxygenase peroxidizes arachidonic acid to
form 5-HPETE, which is converted to its epoxide derivative leukotriene
A4
(LTA4).
LTA4 may be
subsequently hydrolyzed to LTB4 or may be converted to LTC4. 5-Lipoxygenase products influence a
variety of biological processes. LTB4 is a potent chemotactic agent for
polymorphonuclear leukocytes, whereas LTC4 and its hydrolysis product, LTD4, are potent
bronchoconstrictors and mediators of allergic hypersensitivity. These products
are also increased in hypersensitivity diseases such as asthma and thus may have
significant roles in the pathology of such diseases. 5-Lipoxygenase products may
also be involved in cell regulation and maturation as substantial amounts of
5-lipoxygenase are produced when HL-60 leukemic cells are induced to
differentiate. 5-Lipoxygenase represents a key site for the regulation of
numerous biological events, thus the development of inhibitors and the
identification of potential activators of this enzyme are very active areas of
research.

8-Lipoxygenase
The existence of an enzyme that produces 8-HETE in mouse
skin was described by Furstenberger and coworkers in 1986 (1). Brash and
coworkers (2) have recently reported the cloning of this enzyme, which is
strongly induced by topical application of phorbol esters. The cDNA for 8-lox
encodes a 76 kDa protein with approximately 40% homology to most lipoxygenases.
However it exhibits 76% homology to a recently cloned 15-lipoxygenase isoform
expressed in some human tissues (3). The physiologic and pathophysiologic
roles of this enzyme, which are also detectable in brain, remain to be
determined.

This large fragment of the cDNA encoding murine 8-lipoxygenase specifically hybridizes to mRNA
encoding this enzyme, but not with mRNA encoding 5-lipoxygenase, 12-lipoxygenase
or 15 lipoxygenase.

Anti-8-Lipoxygenase (Polyclonal)

12-Lipoxygenase
Three forms of 12-lipoxygenase have been identified,
"leukocyte-type" and "platelet-type" (12(S) LOXs), and a 12(R) LOX in skin.
These differ in tissue localization, substrate specificities, immunogenicities
and sequence homology. The leukocyte-type 12-lipoxygenases have also been found
in other tissues, including the rat pineal gland and human adrenal gland. The
platelet-type 12-lipoxygenases have thus far been found only in platelets and
human epidermal cells. Because of the significant regulatory effects of
12(S)-HPETE and 12(S)-HETE, therapeutic agents which either activate or inhibit
12-lipoxygenase may be useful in treatment of artherosclerosis, hypertensive
disorders, diabetes, and metastatic diseases. Both isoforms of 12-lipoxygenase
have recently been cloned. (Chen, et al., J. Biol. Chem.

12-Lipoxygenase
(Platelet-Type)
This
partially purified preparation of native human platelet 12-lipoxygenase has
negligible enzymatic activity, but is useful as a standard for Western blotting.
Each vial contains sufficient enzyme for at least 10 Western blots.

12-Lipoxygenase
(Leukocyte-Type)
This
stabilized and partially purified porcine leukocyte enzyme has a specific
activity of approximately 50 units/mg protein. The enzyme is suspended in 20 mM
Tris pH 7.2 containing 0.5 mM EDTA, PMSF, DTT, and40% glycerol. Stable at -70°C.
Labile to freezing and thawing. One unit is the amount of enzyme that catalyzes
the conversion of one nanomole of arachidonic acid to 12-HPETE during a minute

15-Lipoxygenase
The enzyme 15-lipoxygenase produces 15-HETE, which can be
metabolized to the trihydroxy compounds lipoxin A4 (LXA4) and lipoxin B4 (LXB4), as well as 5(S),15(S)-DiHETE. The
lipoxins exhibit a range of biological activities. LXA4 has been reported to increase kidney
glomerular filtration rate and to provoke contraction of guinea pig lung. Two
15-lipoxygenase enzymes have been cloned and characterized from human tissues
and are differentially expressed. One isoform (15-LOX form-1) is expressed in a
variety of cell types including neutrophils, eosinophils and macrophages.
Expression of the second isoform (15-LOX form-2), which was identified and
cloned very recently (Brash, Boeglin, et al., Proc. Natl. Acad. Sci (USA)
94:6148-6152, 1997.), appears restricted to lung, hair roots, prostate,
and cornea.

In general, eicosanoids must first be extracted from
serum, using either liquid or solid phase extraction techniques, prior to
immunoassay. In contrast, levels of eicosanoids in urine and tissue culture
supernatant may often be assayed directly following dilution with buffer.

each kit. The following extraction procedure is recommended, for example, prior
to the analysis of prostaglandin E2 in complex biological samples.

15-Epi-Lipoxin A4 EIA Kit
Lipoxins are among a relatively new class of eicosanoids that are
derived from arachidonate and known to be prevalent in airway tissues of
asthmatics and other lung disease patients. This demonstrates that lipoxins are
naturally occurring molecules and closely associated with eosinophil-related
pathways. While their exact roles are not fully understood, both Lipoxin A4
(LXA4) and15-Epi-Lipoxin A4 (15-Epi-LXA4) have proven to be involved in
anti-inflammatory responses. The LXA4, is however, not effective in abolishing
allergen induced eotaxin while the analog 15-Epi-Lipoxin A4 has been shown to be
effective. Furthermore, the 15-Epi-LXA4 has been established as an inhibitor of
eosinophil localization in allergen sensitized tissue making it a molecule of
great interest for tissue inflammation studies. This ELISA kit was
developed to be sensitive and selective for 15-Epi-LXA4 to provide the user a
convenient means to study anti-inflammatory responses. The 15-Epi-LXA4 assay kit
allows for quantitative results with the ease and convenience of a 96 well
format. This assay is a competitive ELISA between enzyme conjugate and
15-Epi-LXA4 for a limited number of binding sites. Our specially designed
extraction buffer eliminates the need for most urine and tissue culture

Lipoxin A4 EIA Kit
Lipoxin A4 (LXA4) is a biologically active product of the
lipoxygenase pathway. LXA4 stimulates leukocyte chemotaxis without aggregation and inhibits
natural killer cell cytotoxicity. It also provokes contraction of parachymal
strips and stimulates microvascular changes. Recent findings indicate that
LXA4 inhibits
