Histamine EIA Kit

Histamine EIA: This heterocyclic primary amine is derived form the decarboxylation of histidine. Histamine serves as a mediator of inflammation and hypersensitivity (anphylaxis) responses. Once sysnthesized by the histidine carboxylase enzyme , histamine is typically stored in metachromatic granules of basophils and mast cells (granulocytes) for later dispersion. Histamine is involved in a cascade physiological effects that are influenced by typical immune response and environmental factors. This assy, like many of our other assays, is developed for quantitative detection while providing both ease and convenience. The histamine assay is a competative ELISA that allows for competition between free histamine from the sample and HRP labeled histamine provided in the kit. A reaction with TMB substrate produces color developement that is inversely proportional to the amount of histamine present. This color can be read at 650 nm or quenched in acid solution to read at 450 nm.

Anti-Human bcl-2 (Sheep): Human hematopoetic malignancies are often characterized by gene rearrangemnts that deregulate gene expression. Antibodies to human bcl-2 (B cell leukemia/lymphoma 2) are directed against the gene product isolated from chromosomal breakpoint t (14;18) of bcl-2, which suppresses programmed cell death (apoptosis) and appears to be modulated by association with bax, a cell death promoter. Recombinant Oncogene Products -

Activation of cellular oncogenes may be directly associated with the development of neoplasia in laboratory animals and humans. Increased expression or structural alterations in these critical genes have been demonstrated to be associated with malignant transformations. Specific methods for the detection of oncogene induction, activation and/or transformation are of great interest. We will continue to expand our products in this area.

 

H-Ras, Wild Type, Recombinant Human: The Ras oncogenes encode four closely related 21 kDa proteins (H-Ras, K-Ras4A, K-Ras4B and N-Ras). The frequent detection of Ras oncogene products in human tumors, strongly implicate a role for these proteins in human tumorigenesis. Ras is a G protein which functions in GDP/GTP regulated signal transduction reactions, including the MAP kinase cascade which activates nuclear transcription factors. This recombinant Ras protein was produced in E. coli and is over 95% pure. It binds GDP and is a substrate for prenylation by farnesyl transferase.

Probes for Acidic Granules

Recent studies indicate important roles for several subcellular organelles that maintain an acidic pH in the development of cellular response receptor-mediated endocytosis.

DAMP: 3-(2,4-dinitroanilino)-3-amino-N-methyldipropylamine (DAMP), MW = 311.3, concentrates in acidic compartments of cultured cells, where it can be visualized after fixation and reaction with mouse monoclonal antibody directed against DNP (product D 07) followed by secondary antibody coupled to an enzyme or fluorescent dye. Useful for localization of organelles where receptor-ligand complexes are targeted, and identification of organelle functions induced by receptor-ligand interactions.

DAMP Kit: A convenient reagent kit designed for cytochemical visualization of acidic compartments. This method can be used in conjunction with light or electron microscopy and offers important advantages over the use of vital pH indicators The unique reagents required for this procedure are: 3-(2,4-dinitroanilino)-3'-amino-N-methyldipropylamine or DAMP (product D 05), a basic congener of dinitrophenol (DNP), and monoclonal anti-DNP (product D 07). Anti-DNP Monoclonal: In addition to its use in conjunction with DAMP for detection of acidic granules (product D 05), this product can also be used for detection of a variety of sparse antigens using immunogold labeling. Detection of sparse antigens with monoclonal anti-DNP involves an antibody "sandwich" technique. Prior to preparation of the sample for electron microscopy, tissue or cells (acting as antigen), are incubated with the antibody. The secondary antibody has been coupled to dinitrophenol (DNP) by a straightforward process, so that it is now labeled with multiple DNP residues. The tissue is then processed for electron microscopy. The monoclonal anti-DNP (the tertiary antibody) is then applied to thin sections followed by protein A-gold labeling. This method has been used to detect the receptor for low-density lipoprotein on human fibroblasts, which had previously evaded detection by protein A-gold in ultra-thin cryosections.

Apolipoproteins

Apolipoproteins have long been known to be ligands for the low density lipoprotein (LDL) receptor. Apolipoprotein E (Apo E) also binds to the b-amyloid and tau receptors, and has been shown to be involved in nerve degeneration and regeneration. Recent studies have found an association of apolipoprotein E with the onset of Alzheimer's disease in late onset families.

 

Apolipoprotein E2, Recombinant Human: This 34 kDa protein has a pI of approximately 6.25, produced in a baculovirus/insect cell system, and purified by affinity chromatography. It competes with iodinated human LDL for binding to the human LDL receptor, and binds to b-amyloid peptides and tau protein in an overlay ligand blot assay. Self association may occur at high Apo E concentrations.

Plasma cortisol levels are useful in the diagnosis of Cushing's syndrome, Addison's disease, adrenal tumors, prostate cancer and depression.

Cortcosterone EIA: This glucocorticoid is secreted by the adrenal gland in response to stress for mammalian species. Glucocorticoids also serve a function in the metabolism of fats, protein and carbohydrate and can be adminstered as an anti-inflamitory agent. Levels of both cortisol and corticosterone increase in mammalian species due to stressors. Only the corticosterone is the dominant glucocorticoid elevated in response to stress for both mice and rats as well as other non-human mammalian species. The actual stressors are not always indicative for the elevated levels of corticosterone but among them are climate changes, changes in environment, rapid temperature changes and changes in daily activity.

This ELISA for was developed for the quantitation of corticosterone in relation to stressors on a test subject. This ELISA is specific for corticosterone with minimal cross reactivity from other key glucocorticoids. Measurement of cortcosterone is obtained with a competitive ELISA design. The corticosterone from the sample competes for a limited number of binding sites with the enzye conjugate. Readings can be taken at 650 nm or 450 nm with stop solution. Oxford provides a specially formulated extraction buffer that allows most sample types to be assayed directly.

Estriol EIA Kit: Measurement of the total estriol has been shown to be useful in monitoring fetal function. Androstenedione EIA Kit: Androstenedione is synthesized in the adrenal gland. It is the primary precursor of testosterone in women. Therefore, in women, androstenedione contributes significantly to the level of androgenic activity, while in men this contribution is negligible. Quantitation of androstenedione may indicate the androgenic activity in women.
