Ecotin: Ecotin is a serine protease inhibitor that is found in the periplasm of Escherichia coli. It has a very broad range of inhibition including the pancreatic serine proteases trypsin and chymotrypsin, human leukocyte and porcine elastase and rat mast cell chymase. Ecotin is regarded as a potent, tight-binding, reversible inhibitor of Factor Xa. PAI-1 Substrates

 

Plasminogen Activator Inhibitor 1 (PAI-1) is a glycoprotein found in plasma that is implicated in the processes of angoigenisis, hemostasis, tumor metastasis, fibrionlysis, ovulation, embryonic development and inflammation (1). The elevation of PAI-1 has shown to correlate with the prognosis of many cancers including breast cancer (2,3,4). PAI-1 is activated by the glycoprotein vitronectin. Vitronectin binds to PAI-1 and modulates fibrinolysis and cell migration (5). This binding has proven to be an important inhibitor of the activated form of protein C (APC) (6).

PAI-1 is a member of the SERine Protease INhibitor superfamily (SERPINS) and will spontaneously decay to the latent or inactive form. This inactive form has a half-life of approximately 1 hour at physiogical pH and temperature. The binding of vitronectin also serves to stabilize PAI-1, nearly doubling its half-life at physiological pH and temperature.

PAI-1 is also found to interact with low-density lipoproteins (LDL) including LDL receptor related protein (LRP), very low-density lipoprotein (VLDL) and gp330/megalin (7).

Human PAI-1 (P9-NBD/ Ser338): The P9-NBD PAI-1 is created via mutagenesis of the P-9 serine residue (Ser338) on the react center loop to cysteine. This promoted the availabilty of a free thiol group for incorporation of N,N'-dimethyl-N- (acetyl)-N'- (7-nitrobenz- 2-oxa-1,3- diazol-4-yl) (NBD), a fluorescent probe highly sensitive to changes in solvation and hydrophobic environment. The fluorescence emission of P9-NBD PAI-1 is enhanced 6-7 fold upon insertion of the reactive center loop into beta-sheet A following complex formation with proteinases, formation of the latent species, or cleavage by elastase. The incorporated probe is excited at 480 nm and displays a broad emission spectrum with a peak centered 542 nm with a resultant blue-shift to 520 nm following reactive center loop insertion. The modified PAI-1 is nearly as active as wt PAI-1 and is more resistant to the spontaneous latency reaction making this an excellent tool for monitoring reaction rates of PAI-1 (1). P9-NBD PAI-1 has been utilized in a number of studies to determine the rates of loop insertion and SERPIN reaction mechanisms when reacted with various proteinases (1,2), inactivating antibodies (3) and conformational changes imposed by the binding of vitronectin (4).

Plasminogen -Sustrates

 

Plasminogen, a single chain glycoprotein zymogen, is the precursor the fibrinolytic enzyme plasmin. The native form of plasminogen is composed of 791 amino acids with glutamic acid located at the N-terminal portion (Glu-plasminogen). Plasminogen has five homologous regions known as kringles. These regions are specific for the complimenting kringles located on tPA, uPA and prothombin. These krigles have one high affinity and four low affinity lysine binding sites that mediate the interactions of plasminogen with fibrin and alpha-2-antiplasm.

Plasminogen is converted to plasmin via a cascade of various reactions that result in the hydrolysis of Arg560-Val561 peptide bond of plasminogen resulting in two chains that remain covalently associated by a disulfide bond.

uPA Antibodies

Urinary-type plasminogen activator (uPA) reacts with many synthetic substrates but the only physiological substrate is plasminogen. uPA¡¯s role in fibrinolysis is to convert plasminogen to the active form plasmin. uPA is inhibited by the presence of PAI-1 which is responsible for the regulation of fibrinolysis.

The uPA receptor (uPAR) enhances the efficiency of plasmin activation and therefore promotes tumor metastasis and invasion. The uPA is furthermore involved in the proteolytic cascade of matrix metalloproteinases (NMP) that degrade collagen.
